Présentation

Le but de mes projets est de réaliser des études fonctionnelles pour comprendre la pathogénicité des variants RyR par l'identification des voies cellulaires responsables de ce processus. Ce projet est directement lié à l'activité de la plateforme Fly Facility.

Recherche

L’objectif de mon projet est une génération de modèles de drosophile (via des méthodes d’édition du génome CRISPR) de variantes génétiques rares identifiées chez des patients atteints de maladies neuromusculaires jusqu’à présent non diagnostiquées. Nos collaborateurs des centres de référence maladies rares de Grenoble et de Lyon avaient sélectionné plusieurs variants de RYR1 codant pour le récepteur de la Ryanodine et également DAG 1 codant pour le dystroglycane. Au cours du projet, nous avons généré trois différentes lignées mutantes stables dans le gène RYR1. Le premier variant pMet4881>IIE variant est situé dans la région formant le pore calcique de RYR1. La deuxième variante IIE3345>F est située dans la région responsable de la régulation du flux d’unités de calcium. Au cours du programme Polonium, nous avons caractérisé avec succès les phénotypes de ces mutants. Deux axes principaux ont été choisis. Le premier est morphologique avec la caractérisation de la structure musculaire, par différentes techniques d’imagerie (microscopie confocale et microscopie électronique). Le second est fonctionnel avec l’étude de la locomotion larvaire et adulte (vol et marche). Les techniques sélectionnées nous ont permis de confirmer la pathogenèse variante. La troisième mutation est le contrôle positif. La mutation Arg R614 >Cys est clairement définie comme pathogène chez le patient. La lignée a été générée et nous commençons à réaliser des tests fonctionnels sur celle-ci. Nous venons de commencer l’analyse fonctionnelle de ces lignes. Il a également été réalisé la planification de la présélection de deux nouveaux variants non diagnostiqués par des généticiens grenoblois qui seront également mis en œuvre en coopération avec le laboratoire polonais.
